QB3/GE Bioprocessing Course

This intensive course on understanding and developing protein purification strategies provides training in theory and practical laboratory skills to members of the biotech industry and academic community. Participants will be trained in the principles and the challenges of developing strategies for separating and purifying proteins, recombinant DNA products and monoclonal antibodies. Hands-on laboratory practicals include product capture, membrane filtration methods, multiple chromatographic techniques and column packing. The training course utilizes state-of-the-art ÄKTA explorer™ platforms driven by UNICORN™ System Control software. The practical sessions allow participants to integrate theory with practice to perform purification of relevant biotechnology products. Formal training certification is awarded upon successful completion of the course.

Location: UCSF Mission Bay, in Byers Hall 212 and Genentech Hall S227.

Fee: Registration is $2,250 for commercial participants; $1200 for academics not in the University of California system; and $200 for UC researchers.

  • Gel filtration

Laboratory:

  • AKTA/UNICORN training

Day Two – Tuesday: Start 8:30

Lecture:

  • Ion exchange
  • Hydrophobic interaction/reverse phase
  • Affinity chromatography

Laboratory:

  • AKTA overview and method writing
  • Gel filtration (buffer exchange)

Day Three – Wednesday: Start 8:30

Lecture:

  • Cell culture/WAVE introduction
  • Membrane separation: introduction
  • Optimization of chromatographic separation

Laboratory:

  • Ion exchange: bead size scouting
  • Optimization of gradient
  • Binding capacity

Day Four – Thursday: Start 8:30

Lecture:

  • Optimization of chromatographic separation (continued)
  • Purification strategies
  • Column packing

Laboratory:

  • Hydrophobic interaction: ligand scouting
  • Salt concentration optimization

Day Five – Friday: Start 8:30 (End 4:00)

Lecture:

  • Scale-up consideration
  • Cleaning of chromatographic media
  • Summary

Laboratory:

  • Column packing and evaluation
